INTENDED USE
Avian flu is high deadly contagion, with the symptom of acute
septic death. The Avian Influenza Virus H5 (AIV H5) Antigen Rapid
Test Cassette (Laryngotracheal secretion/Feces) is used for
qualitative detection of avian influenza H5 virus in avian trachea
and/or feces as an aid in diagnosis of avian influenza H5 virus
infection, such as chicken, bird and duck.
PRINCIPLE
The Avian Influenza Virus H5 (AIV H5) Antigen Rapid Test Cassette
(Laryngotracheal secretion/Feces) is a qualitative, lateral flow
immunoassay for the detection of avian influenza H5 Capsid Protein
HA in laryngotracheal secretion or fecal specimens. In this test,
antibody specific to the avian influenza H5 Capsid Protein HA is
coated on the test line region of the test. During testing, the
extracted specimen reacts with the antibody to avian influenza H5
that are coated onto particles. The mixture migrates up the
membrane to react with the antibody to avian influenza H5 on the
membrane and generate one colored linein the test region. The
presence of this colored line in test region indicates a positive
result. To serve as a procedural control, a colored line will
always appear in the control region if the test has performed
properly.
STORAGE AND STABILITY
Store as packaged in the sealed pouch either at room temperature or
refrigerated (2-30 °C). The test is stable through the expiration
date printed on the sealed pouch. The test cassette must remain in
the sealed pouch until use. DO NOT FREEZE. Do not use beyond the
expiration date.
PRECAUTIONS
Do not use after the expiration date. Do not reuse the test kit.
Handle all specimens as if they contain infectious agents.
Observe established precautions against microbiological hazards
throughout testing and follow the standard procedures for proper
disposal of specimens.
Wear disposable gloves and eye protection when specimens are
being tested.
Humidity and temperature can adversely affect results.
Do not remove test cassette from its pouch until immediately
before use.
Do not mix components from different lot and different products.

DIRECTIONS FOR USE
Allow the test, specimen, buffer and/or controls to equilibrate to
room temperature (15-30 °C) prior to testing.
1. Collect fresh poultry laryngotracheal secretions or feces sample
approximately mwith the swabs provided.
For laryngotracheal secretions sample, make the swab wet
sufficiently.
For fecal sample, the amount of fecal swab as follows:
Note: sample collection directly affects the correctness of test
results, so please ensure proper collection amount.
2. Insert the wet swab into the provided plastic tube with buffer.
Rotate the swab for approximately 10 seconds while pressing the
head against the inside of the tube to release the antigen in the
swab. Remove the swab while squeezing the swab head against the
inside of the tube as you remove it to expel as much liquid as
possible from the swab. Discard the swab in accordance with your
biohazard waste disposal protocol.
3. Place the test cassette on a clean and level surface. Hold the
dropper vertically and transfer 3 drops of extracted sample
(approximately 120 μL) to the specimen well (S) of the test
cassette, and then start the timer. See the
illustration below.
4. Read the result at 15 minutes. Do not interpret results after 20
minutes.
